Currently I am playing the everloving shit out of Spelunky on my Vita (and PC when I have the time) and it's the best thing since toasted bread !

I'd go with The Binding of Isaac. Close seconds is Awesomenauts, especially with the tons of free content they added to the game since release. I really like Hotline Miami, Bastion and Monaco: What's Yours is Mine too!
