Does it work? Yes.
Is it fast? No. Between the fact that only a couple of the highest ranked Support members can take those tickets, and that they are considered low priority compared to other tickets they have to deal with, those often wait a rather long time for a response from Support.

About the rep situation. I just check those fake trade feedback and if I'm going to trade with you I will not mind because the -reps are from users which are not trustworthy. Like one guy is 0/-1 and the other is +1/-3. If they were like +300/0 and they leave -rep on your profile I will check everything before trading with you. I might even add the -rep feedback leaver on steam for more detailed info.

Precisely, that negative rep comes from a (now) suspended user who had a multiple win in the same day...

it will take a lot of time. combine that only some support users can do it and they need to check all the proof you provide, it's not as easy as a reroll.

I had negative rep once, a guy blackmailed me into giving him a free game. Little did he know that Dota 2 would become a free game, that the beta invite was worthless by that point and that I was actually blackmailing him into a permanent ban by landing him the game, making him remove the fake rep and recoding everything he said.

A long time ago I was impersonated and people scammed by the fucker left a couple of negative feedbacks on my profile. I asked support to remove them and they got it done quickly and without much hassle.

The report I filed for your -rep on my profile had been open for like 6 months without a response, then we talked about it again and I closed it.. So from my experience: it takes a looong time. But I indeed have seen people got -rep removed, at least 2 people that I've spoken to (and that had multiple negative feedback which was obviously fake).

I'll share a story with you guys, unrelated.
Back when I was a kid, I used to have a hamster, I called him hamtaro. I fed him and made sure he had something to cover himself at night in his cage, and more hamster stuff. He died like three weeks later.... It was quite a sad time, as a 11 years old dog my mom had died too.
