Bloodlines is great, but I think that Redemption was also a surprisingly fantastic game. Both have huge flaws, so if you're into games which try to do something and don't quite manage to do it right, play Bloodlines. If you want a slightly more polished, safe, and "standard" experience, play Redemption.

Or play both. You know you want to.

I would think the Legacy of Kain games, both blood omen and soul reaver even though in soul reaver you arent really a vampire anymore.

Vampire the masquerade redemption

Vampire: The Masquerade - Bloodlines is the easy answer for me. Feels like Deus Ex in so many ways. Obviously quite a few flaws that the community patch did fix the majority of. Not only do I consider it the best vampire game of all time, it's probably in my top 10 single player games of all time.

I am looking forward to DARK though!! :) Comes out this Friday! Looks like a lot of fun.

Before I finished reading your criteria, I wanted to say Castlevania: Symphony of the Night. Now I don't know. Vampire: The Masquerade Bloodlines is good. Some of the Legacy of Kain games are somewhat good.

As played out as Vampires are on TV and in movies, I just don't think they're really that present in games.
