check device manager to see if everything is working there.
if cable is plugged in properly.
when you click on the speaker button, do you see green bars moving in whatever you trying to play audio?
could try a system restore first to an earlier date when you had sound.

Plugging the cable even from the headphones in the wrong hole, mistake people easily make.
And you are sure your headphones are not broken?
So it's not a fresh install and you had sound that suddenly stopped working? i suggest a system restore.

For future reference there's a program called Audioswitch that lets you change your playback devices if you have more than one. It's very useful. Sometimes a 360 controller gets read as an audio device, or some program hijacks the sound, or it defaults to the other device. You just never know.

If it is a desktop, try plugging into the back instead of the front panel after you've tried all the other good suggestions here.
