Good call using a gift link so that you can tell whether or not the winner redeemed the link. Your best bet is to open a support ticket and request support mark the game received even though the user did not redeem it. The redeemed gift link is a clear indicator that they received the game, even if they didn't redeem the Steam key. The user will likely receive a probationary suspension for failing to redeem the key and leave feedback.
Note, you need to wait 7 days from the time the giveaway ended before submitting the ticket.

Sometimes tickets can take weeks to be resolved. There's a lot of support requests, very few moderators to go through them. And I think they prioritize reroll requests over everything else.

That's a User Report ticket. Make a ticket for "Request Received Feedback" as Semeicardia metioned here. Include a link to your User Report ticket so we don't end up having to deal with this issue twice.
As for the gift, do as Mihey3Z says and ask the game reseller to reissue the key. I advise NOT giving out the new link until you are in direct contact with the person to whom you are giving it. (You can screenshot the conversation when you hand it over and they redeem it.) See Kiva029's post for further details about HB gift links.

Contact humble support, they will say e-mail of someone who revealed your gift link. Also they will generate new gift link. If e-mail is the same as u see on winners page - great ʕ•ᴥ•ʔ If its different - send new gift link again.
If your winner marked as Not recieved after 7 days you should make ticket to support - Misuse of Giveaway Feedback. Permanent suspension.

I just read another topic involving Humble Bundle gift links, so I'm going to mention in case it's relevant:
It's recommended to not generate gift links on Humble until the last moment. Otherwise, they can get incorrectly redeemed: " So, don't generate gift links until the last moment, when you're going to send them. If you generate a link and try to use it after a few days, chances are that the link will be already used (if it's because someone actually got access to the link somehow, or because of a bug in their system that incorrectly marks gift links as used, I don't know)."
So there might be a chance the winner is telling the truth? I recommend doing what Mihey said and see who revealed the link.

I'm pretty sure that support will take care of this (including marking the game as received). The behaviour of the recipient is unacceptable, sorry you had to experience that while trying to do something nice for someone else.

So. Someone won a giveaway I made. After some hours he had seen the key, but hasn't marked as received. Ok. I waited a few days and then I've gone check it on Steam. To my surprise he hasn't activated the game on his account. The key was seen. I clicked on the Humble Bundle gift link. It was redeemed. I contacted him. He claims he hasn't received the key in the first place, says he's reporting me and blocks me on Steam. I reported him, but got no answer from mods.
It got resolved already. I'm waiting on Humble Bundle response now.
