Both games are actually very different.
I´m pretty sure someone already said something similar to this, but anyway i feel like posting.

Dungeon Defenders:
Good game, it has some varied classes but its too focused on grinding gear and stats. The difficulty is mostly gear based.
I did played this game a lot, but then it went the power creep way, where a single item had more stats than your max level character.
Not sure how is the game now, but before playing alone (having 3 or 4 characters) was the best way to beat levels, which was kind of sad.

Orcs Must Die:
Also a good game, the first one doesnt have co-op which was a bit bad, levels are smaller than in DD, its more focused on killing stuff quickly than waiting for hordes of creatures to die in a big level. You do get a lot of traps and weapons to do your job.
It´s much better on the animation part, the character feels fluid (in DD the characters animation are pretty bad and stiff).
It does have less replayability than DD (mostly because of the lack of grinding) but its a great game (i feel bad that i didnt finished the game, but i may do so soon).

Now with Orc must die 2, they seem to have gotten the first game and made everything better, you have Co-op, you can unlock/upgrade your traps, equipment and the like, enviromental hazards to kill stuff and you even get the levels of the first game if you wanted to play those co-op.

Played both, Orcs Must Die is meh.

Dungeon defenders if you have some friends to play with, it just felt like it had much more strategy.
