So I did 3 giveaways over a week ago, which I sent off to all 3 winners (humble bundle gift links via email).
Two have been marked as received, however the 3rd the winner has marked as not received. So today I tried to add that winner on Steam to send them the link to resolve it, however they have rejected the request.

I have checked the gift link and can see they have not claimed it, what is the best course of action here? should I request a new winner since I have made reasonable effort to get this winner their gift?

check his account's games to see if he has the game on his account, he either redeemed it and marked as not received or already had the game and send a support ticket explaining the situation.

It was a DLC, I can see he has the base-game, but I don't believe I can tell if he has the DLC or not?

As said I can tell from the humble bundle gift link that it hasn't been claimed at least.

If it hasn't been claimed, request a new winner then if the winner rejected your communication to resolve your case

request a new winner, but remember to describe whole situation carefully so support could make a right decision. If a game is not claimed (link still works) and the winner doesn't want to talk - i'd concider this as a rejection of a gift.
however be ready to face a situation when the current winner redeem it after all.

It is possible if the winner change mark "not received" to "received" when the OP can communicated with the winner...
