If you mean how giveaways work, once it's over one of the entrants is randomly chosen by the website code as the winner, and you get to see who it is. You then have a week to send the game to them. If you're gifting a Steam key or Humble Bundle giftlink, you can paste it into the site directly; if you're giving away one of the old Steam inventory gifts, from before the change that disabled buying new ones, you can send it to the winner's e-mail, it will be shown to you; if you're buying something from the Steam store to give away, you have to add the winner as a friend on Steam, wait for them to accept, then buy the gift copy and choose them as the recipient.
In any case, once sent, the winner must activate the gift on their account and mark it as received on Steamgifts.

You can make a support ticket, asking the staff to mark the gift received from their end. You have to provide evidence that you sent it, such as a screenshot of the entry on your Steam gifting history (accessible from the inventory) if you sent a gift, or a screenshot of the e-mail if you sent it that way, or a screenshot of the chat session if you sent it that way. If you put a key into the site, I imagine it would be enough to have it there and revealed to the winner, and for them to have left no comments explaining why they didn't mark it as received.
And if they don't accept it in the first place, such as rejecting a Steam store gift, or not viewing a key sent through the site, you can make a support ticket asking for a reroll, meaning a new winner will be chosen randomly.

If you don't provide the game for any reason, the winner will mark it as not received. If you're able to get it later on, you can contact the winner to give it to them and ask them to mark as received. Keep in mind they may already own it at that point, I'm not sure what happens then. In the meantime, having a giveaway marked as not received isn't too major an issue, but doing it multiple times would probably lead to increasingly longer suspensions.
