My first games were Prince of Persia, CD-Man (a colorful Pacman clone), Test Drive II and Tetcolor (Tetris-like game). I played them at my parent's workplace as a pre-schooler and every one of them was too hard for me at the time.
As for Duke Nukem series: I played the first episode of the first game a little later, and it was just the thing I wanted - colorful console-styled arcade platformer with shooting lazers, life counter, robots and explosions, bonuses to collect and all the other stuff. Too bad I played it just once and couldn't find it long afterwards.
When I first played Duke Nukem 3D I had no idea it is somehow connected to that arcade platformer. I was fascinated with interactivity, but it was a little too scary and hard to play on a keyboard. Many years later (when I configurated it to use mouse properly) it became one of my favourite FPS. I finally finished the game when I got the Megaton Edition.
Duke Nukem Forever got me hyped with a demo version, but then the game was released and got trashed by critics, so I finally played it several years later when the storm settled (and GOTY edition key was sold cheap). It was... OK, nothing exceptionally good or bad, often more tedious than fun, the stages without shooting were often more fun that dad guns. I thought it would work better as Postal 2 - style game with lots of civilians and all sort of pranks to do. I actually enjoyed the DLC about Dr. Proton (the arch-nemesis from the first game), again mostly for the non-shooting part.

My first with my first memories goes back to when i was about 2-3 years old playing Bugs Bunny’s Birthday Blowout on the NES. My favourite game of all time is probably Secret of Mana shortly followed by Final Fantasy 7.
For a long time i was a fan of Duke as a character, because i never played any of his games. Only playing some cheap knock off games. DN: Forever was actually the first Duke Nukem Game i ever played and actually wasn't disappointed and enjoyed the silliness of it quite a lot. I recently replayed it to get all the achievements and while i still enjoy it, i also felt it too be little bit too tedious at times and well, its unpolished as hell.

Favourite? I don't know. Never did like picking favourites. Some games I enjoyed a lot were Sentinel on the C64, Planescape: Torment and Peggle, but there were many other games I enjoyed. Some others I remember fondly are HoMM 3, the Monkey Island series and Nine Princes in Amber for the C64. Oh, and UT2003 as the only multiplayer shooter I ever really got into, and City of Heroes was my main MMO.

My first game... can't remember. But it was in the mid 80s, Spectrum 48k. I had a few favourites back then: arkanoid, chuckie egg, manic miner, barbarian, match day and kokotoni wilf (maybe my favourite of all).
Then, came the Commodore Amiga (early 90s I think). Some of my favourite were Kick Off, Rainbow Island and Car Vup.
Jump to PC. Lost count to the games I already played. But if asked to choose a favourite, I'd say Bioshock. Why? One word: Rapture.

I am not really sure, but I think my first encounter with a videogame was some Atari 2600 game but I couldn't tell you which one. Or it may be Space Invaders in Arcade Cabinet in shopping mall but that I didn't play, just saw it, yet it was mesmerizing. And first PC game I played was Supaplex if I recall correctly. I played it back in early 90's in office of major porn publishing company where my father worked. I think I might've been 4 or 5 yo.
About Duke Nukem... I loved both first instalments and 3D as well of course. And I was very pleased with DNF because I wasn't expecting anything else that game which has been in development hell for 15 years and was built on fucking Quake 2 engine. Seriously, what were people expecting? I enjoyed it very much.

I think my first game was Super Mario Bros on the NES. It was a long time ago, so I'm not 100% certain on this, but it's quite likely that it was this one.
Oh and any Duke Nukem fans? I'd love to hear how your first experiences with the original game went, and how bad you felt DN: forever to be.
My first experience with the first Duke Nukem (or Nukum as it was originally called) was not until about 2005. I had already played Duke 3D and Manhattan Project at this point. The first Duke felt a bit. And I have to admit, I did not really like the first Duke Nukem game. It was a typical PC platformer for its era, and I found Commander Keen to be a lot more fun.
Best game to date?
Well, that's not an easy question to anwer. There are plenty of great games out there, and which one I find to be the best depends a lot on my mood:
Planescape: Torment, Europa Universalis IV, Pillars of Eternity, Majesty: The Fantasy Kingdom Sim, Super Metroid, Thief: The Dark Project, System Shock 2, Ultima 7. Vampire The Masquerade: Bloodlines. It's hard to pick one :(

My first game had to be the good old point and click flash games of the 2000s, my most beloved scooby-doo games really helped me get into the Survival Horror genre. First downloadable game had to be CastleMiner Z, better than minecraft, cheeper, and the best on the Xbox 360 to date. First computer game had to be Half Life 2, I adored it at the time. But my favorite game to date has to be Deus Ex: Human Revolution, the atmosphere, the world; everybody loves Skyrim, but in its place I put Deus Ex:HR. What are your firsts and favorites?
